چکیده

Glycerol-fatty acid esterification has been conducted with lipase from R. delemar in water/AOT/isooctane reverse micellar media, with the major product being l-monoglyceride, a useful food-emulsifier. 1,3-Diglyceride was also synthesized, but to a much lesser extent. For a given set of initial conditions, the reaction productivity, measured in terms of the initial product formation rate, Vo, and the final or equilibrium concentration of product, is optimal for a particular concentration of each surfactant, fatty acid, glycerol, and water. Many of these optimal values correlate well with a "critical" region on the phase diagram. Also, results indicate lipase-catalyzed esterification stops due to the achievement of kinetic equilibrium except for a few cases where enzyme deactivation is severe. Dynamic light scattering was employed to examine the influence of water, glycerol, and fatty acid on micellar and interfacial structure. Results from this technique indicate enzyme kinetics are linked to interfacial phenomena and the presence of substrates at the interfacial region.

Lipase Catalyzed Incorporation of Conjugated Linoleic Acid by Transesterification into Sunflower Oil Applying Immobilized Lipase (Lipozyme Thermomyces lanuginosus and Rhizomucor mehei)

Conjugated Linoleic Acid (CLA), Glycerol (G) and sunflower oil blends with varying concentration were subjected to enzymatic esterification using a 1, 3- specific immobilized lipase. CLA was used as acyl due to its purported health benefits. The transesterified lipids were evaluated for free fatty acids (FFA) and composition of fatty acids by gas chromatography. Lipozyme RM IM is preferred for ...

Esterification reactions of lipase in reverse micelles.

The activities of lipase from Candida cylindracea and Rhizopus delemar have been investigated in water/AOT/iso-octane reverse micellar media through the use of two esterification reactions: fatty acid-alcohol esterification and glyceride synthesis. Such media promotes the occurrence of these two lipase-catalyzed reactions due to its low water content. The effect of various parameters on the act...

Lipase applications in food industry

Lipases are the most pliable biocatalyst and bring about a wide range of bioconversion reactions, such as hydrolysis, interesterification, esterification, alcoholysis, acidolysis and aminolysis. Lipases can act on a variety of substrates including natural oils, synthetic triglycerides and esters of fatty acids. They are resistant to solvents and are exploited in a broad spectrum of biotechnolog...

Two-step synthesis of fatty acid ethyl ester from soybean oil catalyzed by Yarrowia lipolytica lipase

BACKGROUND Enzymatic biodiesel production by transesterification in solvent media has been investigated intensively, but glycerol, as a by-product, could block the immobilized enzyme and excess n-hexane, as a solution aid, would reduce the productivity of the enzyme. Esterification, a solvent-free and no-glycerol-release system for biodiesel production, has been developed, and two-step catalysi...

Rapid, sensitive fluorometric determination of serum triglyceride by measuring lipase-liberated fatty acids.

A continuous fluorescence displacement assay was developed for the measurement of long-chain fatty acids and utilized in the study of triglyceride lipase-catalyzed reactions (Wilton, DC. Biochem J 1991;276:129-33). We now describe a method for the rapid measurement of triglyceride in serum with the fluorescence displacement assay. The method involves the hydrolysis of a diluted sample equivalen...
